Sinemet effects with time

I have read in this list and have heard that prolonged use of SINEMET results
in lack of uptake of the vital ingredients in it resulting in odd side effects.
Does anyone know how long, in years, it takes for this to happen? And then what
does one do? Does the concentration of the ingredients affect the time taken
for SINEMET to fail to function as it should. How about a level of 125/100?
Any information on this would be appreciated. Thanks a lot.
